Outline of Annual Research Achievements

①タンパク質アシル化修飾によるロイシン生合成酵素の調節機構
網羅的解析から、高度好熱菌Thermus thermophilusのロイシン生合成酵素IPMDHがその活性発現に重要なリジン残基がスクシニル化修飾を受けることがわかっている。スクシニル化によるIPMDHの活性調節機構についても解析を進めたが、スクシニルCoAによって非酵素的にスクシニル化したIPMDHに活性の低下(変化)がみられなかった。また脱スクシニル化酵素の同定はできたが、脱スクシニル化酵素を欠いた株から調製したIPMDHの活性にも変化が見られず、IPMDHが細胞内でスクシニル化修飾される生理的意義は解明されていない。
②CoA体代謝関連酵素の調節機構
タンパク質アセチル化修飾とNAD+結合タンパク質との相互作用によって活性調節を受けるT. thermophilus由来のCoA transferase(CoAT)について、その活性調節機構の詳細を明らかにするため結晶構造解析を進め、CoATや相互作用タンパク質それぞれの結晶構造の決定に成功した。また、CoATとNAD+結合タンパク質の複合体がNAD+だけでなくNADHも結合し、それぞれに対する親和性が異なることから、細胞内のNAD+/NADH比を感知し、CoAT活性を調節していることを明らかにした。この調節機構が脂肪酸分解経路であるβ酸化への短鎖アシルCoAの供給量を制御し、アシルCoAやNADH生産を通じたエネルギー代謝調節に関わることを提唱した。
さらに、短鎖アシルCoA代謝に関わるアシルCoA合成酵素(ACS)ホモログについても機能構造解析を進め、4つあるACSホモログのうち1つはACS活性を示さないにもかかわらず、ある生育環境では生育に必須なことを破壊株、および相補株の解析から示した。また、本ACSホモログについても結晶構造解析を進め、部分構造の決定に成功した。
