Outline of Final Research Achievements

Skeletal muscle fibers are classified into four types; slow-twitch fibers (type 1), intermediate and fast-twitch fibers (type 2A, 2X and 2B). To date, muscle fiber type composition is thought as an important factor to determine meat quality and nutritional value of meat. However, the relationships between fiber type composition and meat quality have not been fully understood. In this study, we aimed to clarify the gene expression profiling in each fiber type at a single cell level. We succeeded in the single cell RNA-Seq by applying laser microdissection method of a murine muscle cross section. We found the strong correlation between type 2A and type 2B fiber. Moreover, we performed the metabolomic analysis with pork that has different fiber type composition. We revealed that the various metabolites of pork, which may be taste components, are different depending on muscle fiber type composition.

Academic Significance and Societal Importance of the Research Achievements

筋肉には大きくマラソンランナータイプ(遅筋)とスプリンタータイプ(速筋)が存在し、このタイプの違いは我々が口にする食肉の特性にも深く関わると考えられる。本研究の成果により遅筋・速筋の違いにより、味に関与する物質の含有量が異なる事が示された。またこれまでの食肉や筋肉の研究は、筋線維(筋肉の細胞)の集合体である筋組織を用いて検討がなされてきたが、本研究では、筋組織の構成単位である1つ1つの細胞レベルで遺伝子発現プロファイルが異なる事をレーザーマイクロダイセクションと1細胞RNA-Seqという最新の技術を組み合わせる事で達成した。本解析から細胞の遺伝子発現レベルで各タイプ間の類似性が明らかとなった。
