Outline of Final Research Achievements

We established pathophysiological model of glucocorticoid excess using dogs and cultured canine skeletal muscle cells. Glucocorticoid excess significantly induced skeletal muscle atrophy, and increased muscle atrophy-related gene (e.g. GRB10) expressions. The present results suggested that increased catabolism and decreased anabolism of protein through the elevation of these genes might induce glucocorticoid-induced muscle atrophy in dogs.

Academic Significance and Societal Importance of the Research Achievements

グルココルチコイド過剰の生体および細胞モデルが作成されたことで、その病態解明や新規治療法の開発が促進される。そしてグルココルチコイド過剰に伴って生じる遺伝子発現変化などの分子病態の解明により、治療において重要となる標的が明確となってきた。これによってグルココルチコイド過剰による筋萎縮により苦しむ伴侶動物に対する新たな治療法の確立に近づいたと言える。
