Outline of Final Research Achievements |
To examine the role of AhR the cholesterol metabolism, we analyzed the liver AhR specific knockout mice fed a lithogenic diet. The lithogenic diet induced cholesterol gallstone diseases and hypercholesterolemia were prevented in LKO mice. These results revealed that hepatic AhR regulates the excretion of cholesterol and bile acid.

Academic Significance and Societal Importance of the Research Achievements |
近年、一般的な生活を営む人の血中ダイオキシン濃度と糖尿病の発症率との間に正の相関が報告され、低濃度ダイオキシン類の持続的曝露による慢性毒性発現の解明が世界的にも注目されている。しかしながら、これまで低濃度ダイオキシン類の慢性曝露による肝臓の機能異常とそれに伴う肝疾患の発症メカニズムを解明するための有用な戦略がなかったが、本研究で用いた肝臓特異的AhR欠損マウスを用いた解析によりそれを可能にした。本研究より、低濃度ダイオキシンの持続曝露はコレステロール胆石の形成及び高コレステロール血症の発症リスクであることが示唆された。
