Outline of Final Research Achievements

We applied the comprehensive analysis method of mRNA variation in human liver tissues after administration of a drug to chimera mice with highly humanized liver (toxicity of liver examination method:T-LEX method) to a screening search for drug repositioning (DR). We have developed a new candidate drug discovery method for highly effective DR drug discovery. As a result of experiments using disease model animals, we found liver fibrosis-suppressing effect as a new pharmacological effect of an already-approved drug (Drug X) whose safety and pharmacokinetics in humans have been sufficiently proved, and applied for a patent.

Academic Significance and Societal Importance of the Research Achievements

我々が開発したDR創薬の新しい候補医薬品探索法は、探索段階からヒト肝臓組織に対する作用を指標としているため臨床的予測性が高く、ヒト臨床試験での成功率の高い候補医薬品を抽出することができる点、並びに対象因子や対象疾患を事前に設定せず医薬品投与で生じる網羅的なヒト肝臓組織のmRNA変動を調べるため、一回のスクリーニングで対象医薬品の潜在的効果をもれなく調べ複数の新しい薬効を見つけることもできる点が優れており、DR創薬研究分野での学術的意義は大きい。本研究成果は、将来の重大な懸案事項である医療費/薬剤費の高騰を抑えるために効果的な情報を示しており、社会的意義も大きい。
