Outline of Final Research Achievements

Increased expression of mRNA dj-1 has been a common phenomenon in cancers, including breast cancer. DJ-1 protein expression pattern is a prognostic factor and predicts drug sensitivity. To accurately predict prognosis, it is important to strictly control the work process in pathology laboratory to maintain the quality of histopathological specimens. In this study period, the quality of histopathological specimens was improved as a fundamental technology to promote clinical research on DJ-1 and precision medicine. As one of the results, we proposed protocols to prevent cross-contamination.

Academic Significance and Societal Importance of the Research Achievements

酸化ストレス応答を細胞内の高位レベルで制御するDJ-1 は、長期間の酸化ストレス応答過剰亢進状態が続くと、応答システムに破たんを来して癌やパーキンソン病の性格を獲得していく。DJ-1の精密なモニタリングを行うことにより、mRNA dj-1の過剰亢進状態を適切に評価することが実現すれば、癌や痴呆の予防や健康寿命の延長に繋がる可能性がある。そのための社会基盤のひとつとして、病理組織標本作成過程に特殊なポケットを使用することを提唱した。詳細なプロトコールの公表が、DJ-1を含む臨床研究成果の再現性の向上やゲノム医療の確実な実践に繋がるヒントとなることが期待される。
