Outline of Annual Research Achievements

TuD-1細胞株(ラット胸腺髄質上皮細胞由来。JCRB細胞バンク細胞番号:IFO50152)を用いて実験条件の検討を行った。グルココルチコイド(GC)添加群，レプチ ン+GC添加群，レプチン添加群および対照群を作成するにあたり，添加試薬(GCおよびレプチン)の濃度，組み合わせおよび培養期間などについて，各群での培 養細胞の増殖の差が大きく，これらを調整するために各種の条件を検討した。その結果，条件検討にやや時間を要した。 検討結果に即してGC単独添加群，レプチン+GC添加群，レプチン単独添加群と対照群として無添加群を作成した。各群における胸腺間質性リンパ球新生因子 (TSLP)の発現量をELISA法を用いて経時的に測定した。同時にTSLP mRNAの発現量を定量的RT-PCR法を用いて計測した。これらの結果については現在解析を実 施している。 上記と平行して，法医剖検例における胸腺試料を収集し，これらに免疫染色を実施して，胸腺髄質および胸腺皮質中のレプチンレセプターを発現する胸腺上皮細 胞(Leptin Receptor expressing Thymic Epithelial Cells; LR-TECs)を同定した。胸腺ハッサル小体外周を構成する細胞はほぼすべてがレプチンレセプターを発現しており，胸腺ハッサル小体におけるTSLPとLR-TECsの強い関連を疑わせた。胸腺皮質におけるレプチンレセプター発現細胞数を計測したところ，ストレス群では非ストレス群と比較して有意に発現数が減少していた。
これらの成果の一部については国際学会および英語論文の形式ですでに発表した。
