Outline of Final Research Achievements

The aim of this study is to develop a body fluid type-specific genotyping method using each body fluid-specifically (un)methylated CpGs and their neighboring SNPs. We first identified various candidate regions for semen-, blood-, or saliva-specific genotyping using the genome-wide methylation data. We finally demonstrated that the simultaneous detection of methylation status and SNPs in several candidate regions could identify each body fluid-specific SNP type from body fluid mixture samples.

Academic Significance and Societal Importance of the Research Achievements

混合体液試料が現場資料として多く得られる一方で、これらを分離して各体液のDNA型のみ検査をする方法については、物理的に互いの分離が可能な一部の混合体液試料を除いては、実務において有効な検査法はほとんど確立されていない。DNAの状態で混合しているものから、個別の体液DNAを検出する本検査法は、資料の陳旧化によって、物理的分離が困難な場合においても適用可能と考えられ、極めて有効なアプローチとなることが期待できる。
