Outline of Final Research Achievements

By using working heart brainstem preparations from 3 weeks-old Wistar rats, the effects of nociceptin/orphanin FQ on the central respiratory rhythm generation in juvenile mammal (not neonatal) have been examined. Nociceptin/orphanin FQ stopped inspiratory activities in this in vitro model. This suggests that nociceptin/orphanin FQ plays a role as an inhibitory neuromodulator in respiratory rhythm generators of matured mammals.

Academic Significance and Societal Importance of the Research Achievements

3週齢ラットからのworking heart brainstem preparationは、そのviabilityの維持には、標本灌流等の条件の微妙な調整が必要であるが、成熟型呼吸リズム中枢研究の有用なin vitroモデルとなると考えられる。
ノシセプチンが、成熟型呼吸リズム中枢において、呼吸回数を減少させる、あるいは無呼吸を誘発する抑制的神経修飾物質としての役割を示唆するという本研究成果は、ノシセプチンの周術期（麻酔中、後）呼吸抑制への関与の可能性を示唆するものであり、周術期における呼吸中枢と関係する合併症の機序解明、予防、治療に貢献することが期待される。
