Outline of Final Research Achievements	This study revealed that ethanol intensified the production of release of NETs. Addition of ethanol enhanced cfDNA (cfDNA, major component of NETs) release from neutrophils stimulated with PMA. Ethanol also enhanced production of reactive oxygen species (ROS)and citrullinated Histon (Cit-H3), which are essential for NETs production and release respectively. Ethanol alone did not produce NETs, ROS and Cit H3. Polymicrobial sepsis was induced by cecal ligation and puncture (CLP) in acute ethanol intoxication. Plasma levels of cfDNA and Cit-H3 was elevated significantly in ethanol-treated mice compared with control mice.
Academic Significance and Societal Importance of the Research Achievements	急性アルコール中毒時には、敗血症が増悪することが知られているが、その詳細な機構についてはいまだ不明である。今回の研究で、エタノールは、neutrophil exracellular traps (NETs)の産生・放出に影響を及ぼしていることが明らかとなった。NETsは好中球が細菌を捕獲するために放出するDNAやヒストンを主成分をする網目状構造物であり、細菌をトラップすることで感染の進展を抑制する。しかし、過剰な放出は血管内皮細胞傷害などを引き起こし、敗血症増悪に寄与すると考えられる。エタノールが、NETs産生を増大させることが明らかとなり、今後の敗血症治療の発展に寄与すると考えられる。
