Outline of Annual Research Achievements

本研究では研究計画に基づき、「腫瘍分化度と遺伝子変異に基づいた歯原性腫瘍の臨床的遺伝子診断システムの構築」を目的として研究を行った。研究用の症例サンプルは岩手医科大学、広島大学および海外研究協力者から集めた歯原性腫瘍症例（原始性歯原性腫瘍6例、象牙質形成性幻影細胞腫2例、腺腫様歯原性腫瘍8例、エナメル上皮腫2例）を用いた。遺伝子解析には次世代シーケンスおよびPCR法を用い、タンパクの発現解析では免疫組織化学を行った。
その結果、原始性歯原性腫瘍では癌関連151遺伝子と歯の発生に関連する42遺伝子に変異は認められなかった。また正常歯胚と組織形態およびタンパク発現を比較した結果、分化度は帽状期から鐘状期後期に相当すると考えられた。象牙質形成性幻影細胞腫ではCTNNB1遺伝子に特異的な変異が認められた。腺腫様歯原性腫瘍では8例中6例にKRAS変異が認められたが、免疫染色ではMAPK伝達経路でKRASより上流のEGFRも活性化されていたことから、遺伝子変異が腫瘍化にどの程度関与しているか明らかでなかった。
培養実験では、同一患者のエナメル上皮腫組織から初代培養にて樹立されたHAM1～3細胞を用いた。HAM細胞に対して次世代シーケンスによるtranscriptome解析を行った結果、HAM1、3はE-cadherin陽性（vimentin弱陽性）であるのに対しHAM2はE-caderhin陰性（vimentin強陽性）で、HAM2は間葉系細胞の特徴を有していた。しかしながらcell migration assayやwound healing assayにおける細胞の運動能はHAM1、3の方がHAM2よりも高かった。HAM2の間葉系の特徴が由来細胞から受け継いだものか、EMTによるものかは明らかでなかった。
