Outline of Final Research Achievements |
We attempted to get oligodendrocytes using oligodendrocyte precursor cell (OPC)-like cells which is induced from ischemic pericytes. However, it was very difficult to maintain OPC-like cells. In addition, the differentiation efficiency of ischemic pericytes into oligodendrocytes was very low. Ischemic perycytes have limitations in the differentiation ability and in the passage times. Thus we tried to stably maintain ischemic perisytes. As a result, the cells were maintained for long periods of time by the high seeding cell density. In addition, they kept their ability to differentiate. Furthermore, we obtained neural stem cell-like cells by culturing ischemic pericytes with a BMP antagonist.

Academic Significance and Societal Importance of the Research Achievements |
神経幹細胞を得る方法として、発生段階の脳組織から直接得る、ES細胞やiPS細胞の作成を経て神経幹細胞へと誘導する、といった方法が主に知られている。今回、本研究により成体の脳梗塞巣から、神経への分化能を保持し、かつ安定的に長期維持できる神経幹細胞様細胞を得ることに成功した。このことは幹細胞生物学に新たな知見をもたらすだけでなく、脳梗塞などの疾患に対する細胞治療といった臨床応用へつながることが将来的に期待できる。
