The present study demonstrated the effect of leptin stimulation on important reproductive factors and ovarian development in marine teleost chub mackerel (Scomber japonicus). Recombinant leptin was prepared and the following three tests were conducted for functional analysis: (a) in vitro bioassay using primary pituitary cells, (b) chronic leptin treatment for pre-pubertal females, and (c) intracerebroventricular (ICV) administration. Leptin stimulation strongly induced gonadotropin secretion from pituitary cells collected from pre-pubertal females. Chronic leptin treatment significantly promoted ovarian development and triggered pubertal onset. ICV leptin administration significantly upregulated gonadotropin-releasing hormone (GnRH) and gonadotropin synthesis in sexually immature females. These results strongly suggest that leptin is an important signal for reproductive axis activation.
