Outline of Final Research Achievements

Chronic inflammation plays a critical role in pathogenesis of metabolic disorders. Ganglioside GM3, a major molecular species in adipose tissue and human serum, can enhance/inhibit inflammatory processes via an innate immune receptor Toll-like receptor 4 (TLR4) in a acyl-chain structure dependent manner. This project revealed the molecular mechanism by which GM3 species differing in its acyl-chain structures modulate TLR4 activation.

Academic Significance and Societal Importance of the Research Achievements

TLR4は、感染防御において中核的な役割を果たす一方、慢性炎症の要因としても大きな位置を占めており、正と負の側面を併せ持っている。その制御メカニズムの全体像を明らかにすることは、慢性炎症に対する根本的な制御法・治療法の確立につながる。本課題の成果は、体内に存在するGM3の分子構造の違いによって、TLR4が正と負に制御されるメカニズムを示しており、スフィンゴ糖脂質研究の今後の発展に加え、免疫機能や炎症の調節を標的とした医薬開発や臨床診断法への応用が期待できると考えられる。
