Outline of Final Research Achievements |
We performed application of in situ proximal ligation assay to detect the crucial binding of two molecules in malignant tumors. First, we tried to detect the SS18-SSX fusion protein in synovial sarcoma. Subsequently, we detected the binding of tumor suppressor factor WWOX and a small transmembrane protein, TMEM207, in oral squamous cell carcinoma.

Academic Significance and Societal Importance of the Research Achievements |
滑膜肉腫で、SS18-SSX融合遺伝子産物の検出、可視化に成功した。これは染色体転座を有する腫瘍(悪性リンパ腫を含む)で通常ホルマリン固定病理切片で、融合遺伝子産物の特異的検出が可能になる道筋をつけたと考える。また、口腔内癌でWWOXとTMEM207の結合を検出しえたことで、悪性腫瘍細胞での腫瘍抑制因子機能を不活化する異常たんぱく質発現が病理組織切片で検出可能なことを示したと考える。
