Outline of Annual Research Achievements

生体組織を構成する細胞の多くは、日々失われ、新たな細胞へと置き換わる。この仕組みを支えるのが組織幹細胞である。幹細胞は多分化能を保持しつつ増殖できる「自己複製能」を持った特殊な細胞で、多細胞生物の成体においては常に新しい前駆細胞・成熟細胞を供給することで多様な細胞からなる階層性を構築し、これが組織恒常性の維持に寄与する。幹細胞の制御に与る経路については、山中4因子をはじめとする転写因子に加え、近年転写後調節に与るRNA結合タンパクの寄与が注目されている。その一つに細胞運命制御因子MSI2がある。MSI2は、分化誘導因子Numbの発現抑制やアミノ酸代謝酵素BCAT1の発現上昇を介して幹細胞性の維持に寄与し、骨髄性白血病の機能的維持に必須であることをこれまでに私たちは報告してきた。本研究ではこのRNA結合タンパクが細胞内で限定分解を受けること、またこの分解が幹細胞の分化状態に連関していることを新たに見いだした。また質量分析法によりこの分解部位を決定した。さらにリコンビナントMSI2タンパクを用いて切断反応を再現する試験管内反応を構築し、これを用いて切断活性の熱安定性等の性状解析や細胞内局在を明らかにできた。基盤研究Sへの内定に伴って本研究事業を令和3年度で廃止することになったが、今後以上の成果に基づき、切断活性を担う因子の同定と細胞・個体レベルでの機能解析を通じて、幹細胞運命制御の新しい機構を明らかにしていく予定である。
