Research Abstract

近年,細胞のアポトーシス抵抗性獲得が,発がんメカニズムに大きな影響力をもつ経路として重要視されている.そして,がん細胞が獲得した正常細胞にはないアポトーシス抑制機序が,抗がん戦略の重要なターゲットとされてきた.XIAPは,内在性のCaspase阻害因子であり,がん細胞で高発現がみられるアポトーシス抑制因子であることから,最近,新規制がん剤開発のターゲットとして注目されている.
当研究室では,創薬ターゲットタンパク質の標的部位に結合するペプチドを,in silico網羅的スクリーニング手法でデザインし,最適化した結合ペプチドの立体配座に基づくフィルタースクリーニング手法によってミメティクス低分子化合物をデザインする,というコンセプトで創薬を行っている.本研究では,XIAP阻害剤開発の第一段階として,Smac/XIAP複合体3次元構造をもとにXIAP結合能をもつ最適ペプチド配列の決定を行い,in vitro assayでそのXIAP阻害能を検証した.その結果,阻害能が最も高い配列としてAVPFが得られた.このAVPFとXIAP-BIR3との結合pharmacophoreを用いて,Smacミメティクス低分子有機化合物の設計を行い,in vitro assayでIC_<50>数百μMと高濃度ながらもXIAP阻害能を有する化合物を数種見出すことができた.これらの化合物は,これまでに知られているXIAP阻害ペプチドとは全く異なる骨格を有しており,新規低分子阻害剤の開発に重要な意義をもつ.現在,これらの類縁体サーチを行い,構造活性相関を調べることによって,高い阻害能を有するリード化合物の設計を試みている.
