Research Abstract

がん抑制遺伝子によるがん化の場合,染色体の両アレルのがん抑制遺伝子に変異が生じる。2段階モデルの立場に立つと,最初はDNA複製誤りによる片一方のアレルへの突然変異の導入であり,2番目は,Loss of Heterozygosity (LOH)による両方のアレルの失活である。最近の統計によると,がん患者の50%以上にがん抑制遺伝子のLOHが観察されている。DNA複製誤りに伴う突然変異はヒトの場合も,大腸菌の場合も,概ね10^<-7>の頻度である。LOHは染色体全体を標的とする現象なので,その頻度は10^<-3>〜10^<-4>と非常に高い。LOH生成過程のうち染色体喪失(chromosome loss)は染色体不安定性(chromosome instability; CIN)と総称され,mitosisとの関連で生じると考えることが出来る。塩基置換型突然変異は誘発しないが発がん作用のある化学物質を調べ,酵母で染色体喪失を特異的に誘発するphenyl hydroquinone(PHQ)等の化学物質を明らかにした。PHQを用いて細胞周期への影響を調べたところ,Hog1(MAPK)依存的にG2/M境界で細胞周期を止め,高頻度に染色体喪失を導いた。hog1及びG2/M境界の遺伝子swe1遺伝子の破壊株では,染色体喪失は観察されない。またこれらのPHQは,Hog1タンパク質のリン酸化やSwe1タンパク質の安定化に関わり,染色体喪失を誘発することが明らかとなった。ヒト培養細胞をPHQ処理すると,酵母の場合と同様にG2/M期で細胞周期を停止すること,swe1遺伝子を破壊すると,染色体喪失等は観察されない。従って,酵母でもヒトでも同じ機構で,染色体喪失を導くと結論できる。染色体喪失が発がんの重要な要因であることを示している。
