Research Abstract

DNAメチル化をはじめとするエピジェネティクな変化は癌抑制遺伝子の不活性化など癌化において重要な働きをしている。そこで癌におけるメチル化の変化を網羅的に調べることは重要である。我々はMIAMI法と呼ばれるマイクロアレイを用いたメチル化の網羅的解析法を開発してきた。この方法ではメチル化感受性制限酵素Hpa IIと、そのメチル化非感受性のアイソシゾマーのMsp Iを用いてメチル化を正確に検出する。我々は、この方法を用いて肺癌の網羅的解析をおこなった。その結果、未分化型の癌でよりメチル化している遺伝子の多いことや、マイクロRNAをコードする領域の近傍においてメチル化の変化がよくみられることがわかった。また予後不良に関連したメチル化も多数みいだすことができた。それらの中にはEpithelial-Mesenchymal Transition(EMT;上皮間葉移行)に関与するものもいくつか含まれていた。上皮間葉移行とは1980年代初めにElizabeth Hayらが提唱した、上皮細胞が間葉系様細胞に形態変化する現象であり、初期胚発生における原腸陥入、神経提細胞の運動や器官形成過程特に、心臓や腎臓また口蓋形成での重要性がこれまでに明らかとなっている。一方、EMTの獲得が運動性の充進や細胞外基質の蓄積をもたらすことから、癌細胞の浸潤や線維症との関連も示唆されている。今後、これらの遺伝子の肺癌におけるはたらきを明らかにすることにより、治療や診断への発展が期待される。
