Research Abstract

申請者らは家族性大腸腺腫症のモデルマウスである,Minマウスを用いて,その発癌メカニズムを検索してきた。その中で,Minマウス大腸発がんには少なくとも二段階の発がん過程が存在することを報告した(Cancer Res 2002,Cancer Sci 2007)。Apc遺伝子のL0Hはmicroadenoma形成に関与し,発がん初期に重要な役割を果たすことが示唆されたが,一方でmicroadenomaからmacroscopic tumorへの進展メカニズムは未だ不明である。近年我々は,このmicroadenmaからmacroscopic tumorへの進展には,DNAメチル化が重要な役割を果たすことを明らかにしてきた(PNAS2005,MCB 2006)。De novo DNA methyltransferaseであるDnmt3bのコンディショナルノックアウトマウス及びドキシサイクリンによる遺伝子発現マウスを用いた研究より,DNAメチル化の中でも特にde novo DNAメチル化がこの過程に重要であることを明らかにした(MCB 2005,Genes&Dev 2007)。さらに我々は,炎症性大腸発がんのモデルであるDSS投与Minマウスモデルを用いて,globalなDNA低メチル化が炎症発がんに及ぼす影響を検討した。DNA低メチル化は炎症が関与した大腸発癌を著しく抑制した。興味深いことに,DNA低メチル化は炎症による短期的な細胞増殖促進作用には影響を及ぼさないにもかかわらず,持続的な増殖活性を強く抑制した。以上の結果より,DSS誘発Minマウス大腸発がんモデルはDNAメチル化異常の発がんにおける意義を解明するための有用なモデルと考えられ,引き続き解析を行っている。
