Research Abstract

RB1CC1はRBを介した細胞増殖抑制の系とTSC-mTORを介した蛋白合成、細胞成長の制御系をクロストークさせながら、組織、生体の恒常的システム維持に貢献する。本年度は主にRB経路への貢献について、詳細に解析した。
RB経路への貢献は、RB1CC1分子が細胞核内でhSNF5,p53両分子と転写因子複合体を形成することによって、p16,p21,RB1すべての発現を亢進させるものであると明らかにできた。一方で、RB1CC1は細胞質においては、TSC-mTOR経路、mTOR活性化因子として働く。両系は、RB1CC1低発現の骨髄では効率的な細胞増殖継続のために、RB1CC1高発現の神経、筋では過剰な細胞増殖を来すことなぐ、細胞、組織を大型に維持するために機能している。
更に最近、RB1CC1の発現状態が乳癌症例の生存率に大きく関与していることを明らかにできた。RB1CC1の細胞質での働きと細胞核内での機能を病理標本において簡便に判別することによって、臨床的予後が早期に類推でき、治療選択の一指針にもなり得る事実が明らかになった。
