Research Abstract

本研究の目的は,消化管の炎症発がんとその進展における,糖転移酵素発現のエピジェネティックな制御機構を明らかにすることである。消化管上皮細胞ではがん化に伴い種々の糖鎖の発現パターンが変化することが知られているが,その機構は明らかではない。本研究では,正常消化管に発現し,癌転移能を規定する因子の一つであるSd^a糖鎖合成酵素活性が,がん組織において消失するメカニズムとして,糖転移酵素DNAメチル化異常を検証した。
1.消化管炎症(炎症性腸疾患,Helicobacter pylori感染胃炎)における,Sd^a合成酵素のDNAメチル化状態の検討潰瘍性大腸炎26症例,並びにHelicobacter pylori感染胃粘膜30症例(肥厚性胃炎を含む),非感染粘膜20症例の臨床標本を用いて,Sd^a糖鎖合成酵素のDNAメチル化を,COBRA法並びにbisulfite シークエンス法で調べた。その結果,正常粘膜に比べてDNAメチル化レベルが充進していることが明らかになった。とくに潰瘍性大腸炎ではメチル化の頻度が散発性の大腸癌症例における頻度より高かったため,潰瘍性大腸炎に合併した悪性腫瘍での検証をさらに行っている。
2.DNA hypermethylation誘導メカニズムの解析 非炎症粘膜-炎症粘膜-発がんの各段階における糖鎖の発現について,炎症性腸疾患+発癌モデルマウスであるおよび発がん物質(アゾキシメタン)+デキストラン硫酸飲水投与による大腸炎症発癌モデルを用いて検討した.レクチンを行った結果多くのレクチン結合糖鎖は炎症上皮やそれを母地とした腫瘍で発現低下していた.糖転移酵素のmRNAレベルについて検討中を続けている.
