Research Abstract

EBV溶解感染を誘導すると宿主細胞内ではEBVゲノムは100倍から1000倍増幅複製される。このウイルスDNA合成を宿主細胞はDNA損傷即ち異常DNAとみなしATM依存的DNA損傷チェックポイント経路を活性化することを明らかにしてきた。しかしATM依存的DNA損傷チェックポイント経路の活性化はp53の活性化をおこすが,p53及びp53の下流のp21,MDM2の発現量の増加はなく,p53の下流の経路は活性化されていなかった。これはウイルスBZLF1蛋白質がp53をユビキチン化しプロテアソーム依存的に分解することで細胞周期停止や細胞死の誘導をおさえていることがわかった。EBV感染細胞内において,p53は潜伏感染期ではMDM2によって制御されるのに対し,溶解感染期では,MDM2とは別の機構,BZLF1蛋白質自身にはユビキチン活性は無いことから,おそらくBZLF1蛋白質に付随したE3リガーゼ複合体によって制御されていることが明らかとなった。
