Research Abstract

ADAM(a disintegrin and metalloproteinase)は、MMP(matrix metalloproteinase)近縁遺伝子ファミリーに属する分子で、癌細胞増殖・浸潤・転移への関わりが注目されている。本研究課題では、精製したリコンビナントADAM28タンパクを抗原として作製した2種類のモノクローナル抗体を組み合わせてELISA系を開発した。本ELISA系は、MMP-1、MMP-2、MMP-3、MMP-7, MMP-9,MTI-MMP, ADAM8,ADAM9,ADAMTS4とのcross-reactivityがなく、1 ng/mlの検出感度をもってADAM28を特異的に認識することを確認した。肺癌患者(n=102)と健常者(n=12)の血清中のADAM28濃度を測定すると、健常者では平均値が1.1 ng/mlであるのに対し、癌患者では4.5倍有意に高く、病期が上がるにつれて高値を示した(I, II, III, IV期で2.1, 5.1, 10.9, 13.5 ng/ml)。また、腫瘍径が3cm以上の症例はそれ以下の症例より有意に高値であり、リンパ節転移陽性例は陰性例より有意に高値であった。さらに、肺癌切除後1週では術前に比較し有意に低値を示し、再発例では高値を示した。肺癌患者(n=91)のうち75%で尿中に活性型ADAM28がイムノブロット法で検出され、陽性率はI期では24%であるのに対しII-IV期では51%と有意に高く、健常者(n=20)では全てで陰性であった。一方、ADAM28の新規基質探索のため、ADAM28高発現細胞株をADAMインヒビター存在・非存在下で培養し、得られたタンパク質を異なる蛍光色素で標識後、二次元電気泳動法での分離と画像解析ソフトウエアによる定量的差異解析を行い、3種類の新規候補分子を見出した。
