Research Abstract

DNAの静的・動的な変化を総合的に解析し、診断するシステムを作成することは、各人の発がんのリスクと現状を把握する上で極めて重要である。遺伝子上のわずかな違いを長鎖の核酸から検出することはそれぞれの標的に対する極めて厳しい条件の設定が必要であり、高い測定誤差を一元的に解決する系までは到達していない。これからの遺伝子研究で必要なのは、配列の中の見たいところだけを見ることができる手法であり、同時に時間短縮・省力化・高信頼性が期待されている。また、安定したデータを得るために解析の装置化が重要課題であり、メチレーション解析ではデータの定量性も課題になる。したがって、本研究課題では、これら諸問題を解決するための新発想の方法の提案をおこなった。本研究を遂行するために、有機合成化学、光化学、分子生物学の手法を有効にミックスして活用した。発がんに関係する遺伝子多型およびDNAメチル化修飾を効果的に解析するための人工核酸プローブの設計に向けた基礎的な研究やこれまでにわれわれが開発してきた化学的遺伝子多型解析法をシステム化し、産業応用を目指した研究を進めた。具体的には、メチル化をうけたシトシンに対して迅速にオスミウム錯体が形成されることを利用した新規メチルシトシン検出法を開発した。オスミウムとメチルシトシンとの反応を引き起こさせるのに必要なビピリジン配位子を配列認識DNAに取り付けることにより、特定の配列の特定の位置のシトシンでのメチル化の有無に応じて反応を制御することができた。その標的シトシンがメチル化している場合にだけ速やかに錯体を形成するので、短時間のうちに標的シトシンのメチル化量を定量することができた。
