Research Abstract

前年度までのRUNX3ノックアウトマウスや胃癌臨床検体の解析結果に基づき,より詳細なRUNX3の機能解析と遺伝子診断や発癌予防・治療への応用を目指して以下を展開した。
1.胃癌発生母地,特に腸上皮化生や残胃粘膜,残胃癌でのRUNX3の発現変化の検討
2.RUNX3のSNPs(single nucleotide polymorphism)のタイピングと胃癌罹患感受性診断への応用
3.胃癌臨床検体及び周囲背景粘膜における胃癌前駆細胞のスクリーニング
4.胃癌発生リスク評価への応用・胃癌前駆細胞の探索とその追跡調査(コホート研究)
5.複数遺伝子MSPによる遺伝子診断,定量的検査の自動化・血清診断への応用
6.ラット・マウス化学発癌モデルを用いたRUNX3遺伝子発現誘導による発癌予防実験
7.各種悪性腫瘍におけるRUNX3の放射線や抗がん剤感受性への関連の検討
既に我々は、胃癌前駆細胞のマーカーを数種類同定しており、早期多発胃癌切除標本の背景粘膜において胃癌前駆細胞を探索しつつある。また新しいHDAC阻害剤FK228やSAHAなどが開発され臨床応用されつつある。我々はこれらの薬剤がRUNX3を発現誘導し、マウス科学発癌モデルにて発癌抑制効果を示すことを確認しており、RUNX3を分子標的とした発癌予防や癌治療に応用しうると考えている。さらにRUNX3はTGFβ依存性アポプトーシスに重要な役割を果たしており、放射線化学療法の感受性に関与していることが明らかとなった(Oncogene, Sakakura, et. Al., 2007)。また我々はRUNX3ノックアウトマウスとAPCノックアウトマウスの掛け合わせにより大腸病変の発生を確認している。大腸癌の発癌シグナル伝達系(Wntシグナル系)とRUNX3-TGFβシグナル伝達系のオーバーラップする分子も同定されており、今後更なる発展が期待しうる。我々は日立ソフトLuminexシステムにより、多数の検体における複数遺伝子のメチル化を定量的に短時間で測定しうる迅速血清診断システムを確立し、実地臨床に応用可能しつつある。
