Research Abstract

ヘリコバクター・ピロリ菌感染による胃発癌や,肝炎ウイルス感染による肝細胞発癌などに代表される遷延した炎症・感染を基盤とする臓器発癌は,ヒト全発癌要因の約20%を占める。この炎症発癌は,発癌と炎症との因果関係が明確であることから,発癌予防の最も有望な標的となる.報告者は,炎症発癌を起こす動物モデルを作製し,発癌にはβ2インテグリンを介した好中球浸出が必須であること,さらに浸出好中球由来の活性酸素の関与を証明した,これらの成績より,炎症細胞の局所浸出を制御することができれば,炎症発癌を阻止し得るという仮説にたて,浸出阻害に焦点を当てた薬剤探索用in vitroアッセイ系を構築した.これは,96穴プレート上に増殖させTNF-alpha刺激マウス血管内皮細胞上に,マウス腹腔内への異物移入により浸出する炎症細胞を蛍光標識後に重層し,その蛍光量を指標とした血管内皮への接着および浸出の程度を定量解析する系である.がん特定領域・統合がん化学療法基盤情報支援班より提供を受け標準阻害剤(計282種)を解析したところ,34種類に接着抑制効果を,また約10種類程度に促進効果を見いだした,浸出に関わるシグナル候補に下記5系を抽出した.1)ペルオキシソーム増殖剤応答性受容体(PPARs)を介するシグナルで,抗高脂血症薬(ロヴァスタチン)等のペルオキシソーム増殖剤の添加でも同様の結果を得た.2)EGFR,HER2,PDGFRシグナル,3)p38,MAPキナーゼカスケード,4)Akt,PI3K,GSK-3,MDM2シグナル,5)cdk2/9,CDK2/CDKs等の細胞周期制御シグナルである.現在,ペルオキシソーム増殖剤応答性受容体を主な対象として,炎症発癌の動物モデルを用いた発癌抑制効果の検証作業に移行している.
