Research Abstract

腫瘍抗原特異的T細胞のin vitroでの増殖誘導法の確立を目指して、thymic stromal lymphopoietin (TSLP)によるT細胞の増殖誘導機構の解析を平成18年度に引き続き行なった。TSLPにより活性化されたDC (TSLP-DC)では共刺激分子であるOX40リガンドとOX40の発現がともに誘導され、それらの発現がTSLP-DCによるT細胞の増殖誘導に必要であることが明らかとなった。一方、TSLPとToll様受容体(TLR)リガンドにて刺激したDCでは、IL-23の発現が誘導され、その活性化したDCによって、IL-17産生T細胞への分化誘導がおこることも明らかとなった。また、TSLP-DCと自家CD8T細胞の共培養にIL-7を添加すると、増殖誘導が増強され、セントラルメモリーCD8T細胞への分化誘導が促進された。以上から、IL-7添加下での腫瘍抗原ペプチドパルスTSLP-DCまたはOX40リガンド+MHC発現細胞による腫瘍抗原特異的CD8T細胞の増殖誘導が有望と考えられた。
一方、私達は、ヒトTSLPがDC活性化を介してT細胞の分化誘導に重要な役割を果たしていること報告してきたが、まずヒトCD4T細胞において、naiveなT細胞では、TSLPに対する機能的な受容体を発現していないが、T細胞受容体刺激にてTSLP受容体がCD4T細胞に発現誘導され、T細胞の増殖が増強されることを明らかにした。そして、ヒトCD8T細胞においても同様に、T細胞受容体刺激にてTSLP受容体が発現誘導されることを明らかにした。この誘導はCD4T細胞と比較し一過性でありT細胞受容体刺激の強度に依存して生じた。TSLP受容体を発現したCD8T細胞は、TSLPの添加によりDCを介さずとも、抗CD3抗体+抗CD28抗体刺激下において、その増殖誘導が増強された。CMV既感染のHLA-A2402陽性ドナー血単球由来のDCにCMV特異的pp65ペプチドをパルスした場合、IL-2添加と比較して、TSLPを添加することで、非特異的なCD8T細胞の増殖誘導が抑制され、CMV特異的CD8T細胞の増殖を優位に誘導できることが明らかとなり、腫瘍抗原ペプチドを用いた腫瘍抗原特異的CD8T細胞の増殖にも応用できる可能性が示唆された。
