Research Abstract

Epstein-Barr virus(EBV)の核抗原であるEBNA1はEBV陽性癌に共通して発現している蛋白質である。EBNA1を標的抗原として活用できれば,すべてのEBV陽性癌に対して有効な免疫療法が構築できる可能性がある。また,効率的な腫瘍免疫応答には,CD8陽性のCTLのみならず抗原特異的なCD4^+T細胞の動員が重要であることが指摘されている。
遺伝子導入した抗原提示細胞は,通常HLA class I経路によってCTLへ抗原を提示する。導入遺伝子産物をclass I経路のみならず,HLA class II経路にも誘導できれば,CTLと抗原特異的CD4^+T細胞を同時に活性化することが可能になる。Lysosome-associated membrane protein(LAMP)-1,heat shock protein gp96,HLA class II-associated invariant chainあるいはautophagy-related protein light chain 3と,EBNA1との融合蛋白を発現するplasmidをそれぞれ作製し,それぞれをHLA-DR^*A0101とHLA-DR^*B0401と同時に293T細胞に導入し,EBNA1特異的HLA-DR^*0401拘束性CD4^+T細胞クローンを反応細胞としたELISPOT法を行った。この結果,EBNA1蛋白N-末端にheat shock protein gp96のleader sequenceを融合し,EBNA1蛋白C-末端にLAMP-1のtransmembrane domainを融合した構造物が,最も効率よくCD4^+T細胞エピトープを提示できることを明らかにした。今回特定したclass-II targeting構造は他の腫瘍抗原特異的CD4^+T細胞の誘導,解析にも応用可能である。
