Research Abstract

l.CN関連遺伝子のENU変異マウスのスクリーニング
Dynamin、CamKII betaなど5つの候補遺伝子について、理研GSCの協力により「ENU変異マウスライブラリー」のスクリーニングを進め、13領域から11変異が見出された。この中にはアミノ酸置換変異が1つ見出されており、その変異は当該タンパク質のリン酸化特性に関わると考えられる。
2.遺伝子改変マウスの行動解析
CN関連遺伝子の遺伝子改変マウス6系統について網羅的行動解析の結果、個々の行動テストではCNマウスと同方向の表現型(=統合失調症様の表現型)が多数見出された。一方で、逆方向の表現型を示したものもあり、単純な結果にはならなかった。これまでに共同研究を含めて70系統以上の遺伝子改変マウスについて網羅的行動解析を実施しているが、ほとんどの系統でなんらかの行動表現型が見出されるという結果を得ている。この手法をさらに進めていくことにより、遺伝子と行動表現型をつなぐ重要な情報を蓄積できると考えられる。
3.顕著な行動異常を示すENU変異マウスの行動解析
若菜・権藤らによる一次スクリーニングで得られたENU変異マウスについて網羅的行動解析を行った。この変異マウスは活動性の亢進、不安様行動の低下などの精神疾患様の行動異常を示した。その他にもスタートルレスポンスの低下を示した。
