Research Abstract |
ヒトの高次脳機能形成がどのような自然選択によって形作られたかを解明するために、グルタミン酸受容体遺伝子群について以下の分子進化学的解析を行った: 1)グルタミン酸受容体遺伝子の上流調節領域の多型検索 NMDA型5種、AMPA型4種及びカイニン酸型5種の遺伝子の上流領域についてヒト50-72検体、チンパンジー24-50検体での全変異検出と中立性の検定を行った。その結果、GRIN2Dにおいてヒト-チンパンジーに共通した純化選択が強く示唆された。同様にGRIN2B, GRIA4, GRJK3においてチンパンジー特異的な純化選択が強く示唆された。一方各遺伝子のウィンドウ解析では、同じくGRIN2B上流領域においてヒト特異的にTajima's Dが有意に上昇する(+2.16, p<0.05)領域が見出され(転写開始点の上流3.0kb付近)、この領域がヒト特異的平衡選択を受けている可能性が示唆された。 2)グルタミン酸受容体遺伝子群におけるヒト特異的アミノ酸置換の同定 前年度までに同定していた、グルタミン酸受容体全26遺伝子におけるヒト-チンパンジー間のアミノ酸置換74ヶ所について、ゴリラ、オランウータンなどの他の霊長類8種の該当配列を決定した。その結果、チンパンジーとの分岐後ヒト系譜側でおきた塩基置換を41個同定した。さらにそれらを世界各地の7集団からなるヒト80検体でタイピとグし、そのうちの34個がヒトで固定していることを確認した。このうち、12個は重要なモチーフの変化をともなうことを見出し、ヒト系譜において自然選択の対象となった可能性の高い重要な機能変化をもたらしたことがアミノ酸置換であることが示された(論文準備中)。
