比較ゲノム解析に基づくヒトMHC領域の進化形成過程の解明

Research Abstract

本研究課題では、ヒトと比較的近縁である霊長類サルならびに非ほ乳類のMHC領域のゲノム配列を決定すること、得られたゲノム配列と既知のゲノム配列情報を含めて、比較ゲノム解析に必要な情報を抽出することにより、現在のヒトのMHC領域に至るまでの進化形成の過程を明確にすることを目的とした。本年度における成果を以下に記す。
1、4.2MbのマーモセットMHC(Caja)領域を網羅するBACコンティグマップを作成し、HLA領域とのゲノム構造を比較した結果、その構造はHLA領域とよく類似したが、CajaクラスI領域には遺伝子重複により生成された数多くのMHVCクラスI遺伝子が位置した。それらのうち、HLLA-BやHLA-Cの直系遺伝子である10個のCaja-Bは、いずれとも遺伝子発現が認められなかった。
2、オキゴンドウクジラMHC領域2.2Mbのゲノム配列を決定した。この領域に相当するヒトゲノム配列の長さは4.3Mbであることから、オキゴンドウのMHC領域は極めてコンパクトなゲノム構造を有していた。さらに、ほ乳類間の比較ゲノム地図の概要版を完成させた。次の段階として、MHC領域に位置する約140個の遺伝子それぞれにおける機能的モチーフの保存性や進化速度の種間差などを明らかにし、ほ乳類MHC領域の進化形成過程ならびにほ乳類におけるヒトの位置づけを進めている。
3、獲得免疫の起源の解明に最適な動物を特定するために、進化した形態を有するネズミザメ上目20種に、より原始的な形態を有するツノザメ上目10種を含めてシトクロームb遺伝子における系統関係を調べた結果、生きている化石に指定されているラブカ(ツノザメ上目カグラザメ目)は板鰓類の中で最も古くに種分岐した証拠を得た。本年度にMHCを含む獲得免疫の起源やその進化過程を理解するための良質なサンプルを入手したことから、ラブカの遺伝子ライブラリーを作成するための予備実験を進めている。
