Research Abstract

国際コンソーシアムでのプロジェクトを通じて高頻度なCNV領域については当初の想像以上に500Kアレイのプローブが抜け落ちていることが明らかになった。CNVの領域ではアレル数の不均衡によるcall rateの低下、Mendel InheritanceエラーやH-Wエラーによってアレイの開発段階で多くのプローブが抜け落ちていることがわかった。こうした500K arrayではカバーできないCNV領域が多数見られたという事実を踏まえ、CNVカバーのよいより高密度なアレイプラットフォームの開発が必要とおもわれた。我々がAffymetrixと共同で開発したオリゴヌクレオチドアレイは全ゲノムで130万(1.3M)のデータポイントと従来の500K arrayの倍以上の情報が取得可能である。CNV領域のカバー率も良く、次世代ゲノムアレイ SNP6.0 arrayのプロトタイプとなった。このアレイにGIM/GEMCAアルゴリズムを実装した。先天性腫瘍性疾患Gorlin症候群のゲノムDNAの解析ではPTCHを含む最少165Kbpの欠失が見つかりいずれの症例にもbreak point(切断点)が同定可能であった。
