Research Abstract

恐怖や不安記億の再生は交感神経活動の変化を伴うがその詳細は不明である。睡眠REM期には記憶の再生と固定が行われており、睡眠REM期の海馬神経活動と交感神経活動の関係は、記憶の再生と交感神経活動変動との因果関係を検討するモデルとなる。本研究は、睡眠REM期における海馬CA1領域の神経活動と地域血流量の変化の定量化し、睡眠REM期と覚醒運動時の行動ステージとの差を検討し、睡眠REM期の特徴を明らかにすることを目的とした。また、腎および腰部交感神経活動との相関性について検討した。Wistar系の雄ラットを用い、脳波、海馬CA1神経活動、海馬CA1領域血流量、動脈圧、中心静脈圧、心電図、筋電図測定のための電極、プローブ、カテーテルを慢性留置した。海馬CA1神経活動は、4極のステンレススティールMicro-wire arrays電極によリ計測した。海馬CA1神経活動は、REM機が最も低く他の行動ステージに比べて有意に低かった。一方、睡眠REM期の海馬CA1領域血流量は、ノンレム期に比べて増加した。以上、睡眠REM期の海馬CA1神経活動は、睡眠-覚醒の行動サイクルの中で最も低い値を示すが、血流量は最も高い値を示すことが明らかになった。本研究は、睡眠REM期海馬CA1領域の血流量は神経活動低下時に増加していることを明らかにした。脳神経細胞では、一般に神経活動とその領域の血流量は同じ方向に変化する(neuro-vascular coupling)と考えられている。しかし、睡眠REM期にはneuro-vascularのuncouplingが生じている。従って、睡眠REM期には海馬の錐体細胞以外の代謝(グリアなど)の細胞の代謝亢進が考えられる。すなわち、睡眠REM期には記憶情報処理自体の活動が抑制され、その周辺機能の亢進が生じている可能性を示唆する。
