Research Abstract

脳幹は、生命活動に関わる多くの情報が直接入・出力し、処理・統合される重要な領域である。脳幹では、末梢からの情報の伝達回路と、脳幹に内在する自発興奮の伝達回路とが混在しており、これらは互いに影響をおよぼし合いながら機能的に形成・調節されていることが推察される。中枢神経系では、発生過程において、シナプス結合はいったん過形成され、後にrefinementされることが知られているが、この過程に自発興奮が重要な役割を果たしていることが示唆されており、脳幹内神経回路網の機能形成過程を明らかにすることは、中枢神経系における神経回路網形成の基本原理を明らかにすることにつながると考えられる。我々は、発生期の中枢神経系において、脳神経・脊髄神経を介した外来性入力、あるいは中枢神経系内に内在する自発興奮活動によって、大脳から脊髄にいたる中枢神経系のほぼ全領域にわたって広範に伝播する脱分極波(depolarization wave)が誘発され、それに引き続き、Ca waveが引き起こされることを見いだした。この脱分極波は、脳幹において神経核内のシナプス伝達が開始され、神経回路網が成熟していく時期に一致して特異的に出現することから、脳幹回路網形成に重要な役割を果たしていると考えられる。さらに、この脱分極波は、鶏胚に限ってみられる現象ではなく、ラット胚やマウス胚においても観察されることが明らかとなっている。
本年度は、自発性脱分極波の発生段階の早い時期での起源を明らかにすることを目的として、哺乳類と比較して取り扱いが簡単な鶏胚を対象として実験を行った。発生初期の鶏胚中枢神経系標本に光学的イメージング法を適用し、脱分極波の起源を調べたところ、起源はまず上部頸髄に出現し、発生とともに起源の数と位置が多様化することが明らかとなった。
