Research Abstract

本研究の目的は神経因性疼痛モデル動物の痛覚伝導系における接着因子Ll-CAM分子の挙動と疼痛との関連の解明にある。平成17年度末で、LIに関する研究は一段落し、Eur. J. Neurosciに発表した。平成18年度は、脊髄後角の接着因子のダイナミックな変化を惹起する因子として、tPA(tissue type plasminogen activator)に注目し、検討を進めた。
1.神経根損傷に伴うtPAの発現変化の詳細な検討
ラットのL5神経根損傷モデルを作成し、脊髄後角でのtPA発現の経時的な変化を確認し、その変化を定量した。神経根損傷後、1日目に既に大きく増加し、mRNAレベルでは7日目まで、蛋白レベルでは14日目でも増加が継続していた。
2.tPAの発現細胞を明らかにする
脊髄後角において、各種のマーカー蛋白などとtPAを組み合わせた多重染色を行い、レーザー共焦点顕微鏡で観察すると、発現細胞はアストロサイトであることが明らかとなった。
3.脊髄後角におけるプロテアーゼ活性の変化
神経損傷モデルの脊髄後角におけるプロテアーゼ活性の上昇をin situ zymographyで確認出きた。またtPA STOPというtPAの阻害剤を投与すると、その変化が消えることを確認できたので、これらの変化がtPAの変化に由来することがわかった。
4.疼痛行動の計測
神経損傷モデルに対して、tPA STOPを投与すると、機械的刺激に対する過敏反応(mechanical allodynia)の形成を抑制することがわかった。しかし、過敏反応形成後に投与しても効果がないことより、mechanical allodyniaの維持には関与していないと考えられた。
以上の結果は、後根損傷により、脊髄後角表層においてアストロサイトの活性化が起こり、tPAを分泌、その周囲の細胞外マトリクスの修飾や接着因子等の組織分解により、脊髄後角の可塑的変化を引き起こしていると考えることが出来る。この変化が結果的に疼痛行動過敏につながることになると思われる。これらの結果は、まとめてGLIAに発表した。
