Research Abstract

(1)VSOP1について基本構造(ストイキオメトリー)を明らかにする目的でwestern blotによる解析を行った。Heat感受性のダイマーのバンドが見られ,クロスリンカーで明確なdimerの位置にバンドが見られた。共同研究者Peter Larssonと蛍光ラベルによる計測を行ったところ,FRET現象が確認され,オリゴマーであること,とくにdimerである可能性が高いことが明らかになった(PNASにin press)。VSPについては酵素との共役とゲート電流特性を明らかにし(J.Physiol.2007;J.Biol.Chem.In press),基質特異性を明らかにした(PNASにrevise中)。
(2)それぞれの電位センサー蛋白の機能を調節する因子を同定する目的で,想定される分子との共発現による免疫沈降実験,two hybridスクリーニングを行った。現時点では機能的に結合する分子は同定されていないが,条件を変えて検討中である。
(3)崎村教授らによる支援班を通じ,VSOP1のコンディショナルB6ノックアウトマウスの作成が進み,既にES細胞が樹立されたVSOP1についてはキメラマウスを作成中である。
(4)免疫組織学的解析により,成体マウス脳においてミクログリアにおけるVSOP1の発現を確認した。胎児時のミクログリアには,発現が確認されないことから,発生時期依存的な発現を起こす可能性が示唆された。
(5)VSPの電位センサー領域を用いて蛍光タンパクとの融合分子を作製し,膜電位レポーターとして開発する実験を理研の宮脇グループと共同で進め,変化率の著明に高いプローブを得て,現在分子特性とイメージングへの適用について実験を進めている(投稿中)
