Research Abstract

[研究の背景と目的]
DISC1結合タンパクである14-3-3ε遺伝子(YWHAE)の5' flanking regionに位置するSNP-1(G>C)の頻度が統合失調症では有意に少ないことを遺伝統計学的に確認し、さらに、C allele (C/C or C/G)を有する個体はG/Gを有する個体より末梢血において発現が増加していること、14-3-3ε遺伝子のノックアウトマウスは作業記憶の障害、不安様行動の増加が見られた。
以上を踏まえ、14-3-3ε遺伝子のSNP-1が診断的表現型(統合失調症か否か)だけでなく、中間表現型であるMRIによって撮像したヒト中枢神経系の構造に対して、どの様な影響を与えるかを検討した。
[方法]
・対象: 本研究に関して文書で説明し、同意を取得した統合失調症33名と健常者29名
・MRI撮像とVoxel-based morphometry法により、関心領域の体積を測定した.
・対象から得たゲノムを用いて14-3-3ε遺伝子のSNP-1の遺伝子型を決定した。
・遺伝子型が各関心領域の体積に与える影響を、統合失調症および健常者で検討した。
[結果]
・統合失調症患者の海馬体積は、SNP-1のC allele (C/C or C/G)を有する個体がG/Gを有する個体より体積が有意に大きかった。この差異は健常者では見られなかった。
・他の脳部位に関して、統合失調症患者も健常者も遺伝子型による体積の影響を見られなかった。
[考察]
・SNP-1のC alleleは統合失調症発症において予防的に働き、14-3-3εの発現も多く、海馬の構造萎縮にも保護的であると考えられる。
・14-3-3ε遺伝子のgenotypeがMRIで撮像したヒト中枢神経系の構造に対する影響を与ることが確認された。現在、独立した他のサンプルでの確認中である。
