Research Abstract

天然有機化合物、生理活性物質には、多環性複素環骨格を有するものが数多く見られることから、各種の多環性複素環骨格を高効率的に構築可能な反応が開発できれば、それらの合成に有用な手段を提供できることになる。我々は最近、遷移金属錯体により活性化されたアルキンに対するカルボニルあるいはイミンの付加によって、遷移金属部位を含む双性イオン(遷移金属含有1,3-双極子)が生成することを見出している。この活性種は、1,3-双極子とカルベン錯体の双方の性質を持つ新規化学種であり、この特徴を活かした分子設計によって多環性複素環を一挙に構築できる触媒反応を開発することが可能となった。
前年度の検討では、ベンゼン環の隣接位にアルキン部位とカルボニル(またはイミン)部位を導入した反応基質に対して、ビニルエーテルの存在下で触媒量のW(CO)5(thf),またはPtCl2を作用させると、金属含有カルボニルイリド(またはアゾメチンイリド)が室温下で効率よく生成し、これがビニルエーテルと「3+2]付加環化反応を起こすことで含酸素(または窒素)多環性化合物が一段階で収率よく合成できることを明らかとした。本年度はこの反応をより有用性の高いヘテロ環骨格の簡便な構築手法へと発展させることを主目的として検討し、ベンゼン環をもたない鎖状の反応基質を用いても同様の金属含有イリドの生成とその付加環化反応が効率よく進行することを見いだした。さらにここで開発した新反応を利用して生理活性物質の全合成研究も展開した。
