Research Abstract

癌抑制遺伝子p53の標的分子であり,G1期での細胞周期停止に関与するp21の発現誘導剤の探索において,我々は新規ヒストン脱アセチル化酵素(HDAC)阻害剤を発見した。HDAC阻害剤は,現在抗癌剤として極めて強い注目を集めている生理活性物質であるが,今回,我々が見いだしたYM753は,従来のHDAC阻害剤に比べて活性が強く,実際にin vivoでも強い抗腫瘍効果を確認している。しかも正常細胞への毒性が低さやin vivoでの副作用が低いことも見いだしている。
癌抑制遺伝子p16のファミリー遺伝子であり,G1期での細胞周期停止に関与するp15^<INK4>の発現誘導剤の検討において,我々はMEK阻害剤を発見した。MEK阻害剤も,現在抗癌剤候補物質として極めて強い注目を集めている生理活性物質であり,我々が見いだしたJTP-70902は,in vitro及びin vivoでも癌細胞増殖阻害を確認している。
抗腫瘍性リガンドであるTRAILは,現在臨床開発中であるが,そのTRAILの受容体であるDR5発現誘導剤の検討において,カロテノイドであるhalocynthiaxanthin,peridinin,リポキシゲナーゼ阻害剤であるnordihydroguaiaretic acid,抗血小板薬であるdipyridamoleに,それぞれDR5発現誘導能がること,そしてそれらとTRAILとの併用によって,アポトーシスの誘導が,TRAIL単剤に比べて著しく増強することを確認した。
