Research Abstract

システイン(チオラート)と遷移金属の結合部位はタンパク中に広く存在し、電子移動やイオン濃度の制御に関与する、あるいは酵素活性中心として小分子活性化を触媒する。本研究ではシステインを模倣する多座の硫黄配位子を用いて、酵素活性中心にある種々の金属チオラート/スルフィドクラスターを合成化学的に構築し、その性質や反応性から酵素活性部位の構造と機能の本質に迫るべく、特にアセチルCoA合成酵素と[NiFe]ヒドロゲナーゼに注力して研究を進めた。
N_2S_2型配位子を有するニッケル錯体とNiBr_2の反応から得られるNi-Ni-Ni三核錯体に、テトラメチルチオ尿素(tmtu)の存在下、4倍当量のAg(OTf)を反応させ、さらに1倍当量のMeMgC1とKSDmpを順次反応させたところ、一方のNi上にメチル基とチオラートを併せ持つNi-Ni二核錯体が得られた。これは室温で一気圧のCOと速やかに反応し、還元的脱離を経てNi(0)カルボニル錯体と、チオラートがアセチル化されたアセチルCoA類縁体であるCH_3C(0)SDmpを収率80%で与えた。本反応の結果は、実際の酵素活性中心においても、二つあるニッケル中心のうち末端ニッケル側だけで反応が進行することを示唆している。
水素の発生と消費に関わる[NiFe]ヒドロゲナーゼの活性部位を再現する無機合成は困難を極めており、構造と機能の関係に迫る研究は世界的にも進んでいない。我々は、多座チオラートを組み込んだカルボニル/シアノ鉄錯体を鍵前駆体としてFe-Ni二核錯体の合成に取り組んだ。反応条件、特に空気と熱の両方に不安定な化合物の取り扱いについて種々検討し、[NiFe]ヒドロゲナーゼ活性部位の構造を模倣するモデル化合物の合成にも成功した。
