Research Abstract

本申請研究では、ヘムタンパク質のヘムポケットをユニークなナノ配位空間と位置づけて、この柔軟かつ緻密な天然の配位空間内に新しい金属錯体を構築し、得られた生体金属分子について、その錯体物性の制御、反応活性種の形成、さらには人工金属酵素としての応用を図ることを主目的としている。具体的には、本年度もアポ化した(天然のヘムを除去した)タンパク質に対して、反応性に優れた人工ヘムをヘムポケットに挿入することにより、天然を凌駕する酸化酵素の構築を以下のように行った。
1.ヘムオキシゲナーゼの基質ポケットに、分解しない安定な非天然ヘムである鉄ポルフィセンを挿入し、新しい酸化触媒を構築した。さらに、アポ西洋ワサビペルオキシダーゼ(HRP)中にも鉄ポルフィセンを挿入し、鉄ポルフィセンでは、初めて2電子酸化等量のFe(IV)ポルフィセンラジカル種をESRスペクトルで捉えた。またこの再構成HRPは、天然の酵素を凌駕する活性を示した。
2.亜鉛ミオグロビンのタンパク質表面にアニオンドメインを導入し、電子メディエーターのビオロゲンをもちい、光増感剤として利用することを検討した。特に、モノカチオン種のビオロゲン類縁体をメディエーターとして用いることにより、光誘起電子移動後の電荷分離状態の寿命が長くなることが明らかになった。この知見をもとにさらに、系中に白金コロイドを添加し、犠牲試薬としてトリエタノールアミン存在下、効率の良い水素発生系の構築が実現した。
