Research Abstract

本研究では,環境制御型電子顕微鏡下でのナノマニピュレーションシステムに基づいて,ナノツールを単一細胞に対してバイオ応用するための各種技術課題について検討してきた.
本年度は,昨年度より構築してきた環境制御型電子顕微鏡ナノマニピュレータに基づいて,バイオナノツールを利用し,単一細胞の局所的な硬さを計測するための技術について検討した.イースト菌やビブリオ菌などの生体試料を環境制御型電子顕微鏡に導入し,高真空モードと環境制御モードでの菌体の状態を比較した.イースト菌は,高真空モードでは多くの細胞が変形し凹んでいるが,環境制御モードでは多くの細胞が球形の通常の形状を保っていることを確認した.さらに,両観察モードに導入したイースト菌を,導入後,再度プレート培養させた結果,高真空モードに導入した場合と比較すると,環境制御モードでは観察前の通常の培養状態とほぼ同等の多くの細胞コロニーを形成していることを確認した.以上により,環境制御型電子顕微鏡用ナノマニピュレータへの細胞導入の可能性と有効性について検討した.また,環境制御型電子顕微鏡用ナノマニピュレータに原子間力顕微鏡用カンチレバーを取り付け,単一細胞の任意の位置に応力を印加したときの細胞の変形量を画像計測し,局所的な硬さ情報を計測した.細胞に印加した力は,カンチレバーの変形量を画像計測することにより算出した.高真空モードや環境制御モードといった圧力条件,増殖段階や細胞のサイズといった培養条件を変化させたときの硬さ特性を比較評価した.この結果,高真空モードや環境制御モードでは,細胞の硬さに明確な違いが得られた.また,各増殖段階に応じて,カンチレバーが細胞を貫通するための貫通力が増加していくことが明らかとなった.以上により,環境制御型電子顕微鏡ナノマニピュレータによる単一細胞の局所的な硬さ計測を行い,その有効性を確認した.(791字)
