Research Abstract

小胞型グルタミン酸トランスポーター(vesicular glutamate transporter, VGLUT)はグルタミン酸による化学伝達の必須因子である。グルタミン酸作動性神経のシナプス小胞に局在しており、グルタミン酸の小胞内濃縮を司っている。神経がグルタミン酸を分泌するためには、VGLUTがシナプス小胞に特異的に組み込まれることが必要である。しかしながら、その機構は全くわかっていない。本研究により、VGLUTのシナプス小胞へのターゲッティングを分子レベルで解明することに成功した。
(1)VGLUT2のトポロジーを解明した。
(2)精製VGLUTをリポソームに組み込みグルタミン酸輸送活性を再現できた。すなわち、in vitroのVGLUT機能測定系を構築した。この実験系を用いて、様々な変異VGLUTの機能を測定した。これらの変異がターゲッティングに及ぼす影響を現在調べた。輸送機能は維持しているがターゲッティングに変化がある変異体を複数同定した。さらにVGLUT2のカルボキシル末端近傍(少なくとも20アミノ酸残基)がターゲッティングに関係することがわかった。
(3)VGLUTI KO miceを用い、末梢組織におけるグルタミン酸シグナル伝達系の生理的意義を解析した。小腸のL細胞におけるGLT-1内分泌におけるフィードバック制御ならびに骨における骨消化制御を発見した。特に、VGLUT1が osteoclastのtranscytotic vesicleに局在することを発見した。このため、osteoclastsは骨消化の際、骨消化物質とともにグルタミン酸を分泌する。グルタミン酸シグナリングは骨組織の恒常性を支える新しい制御系であると考えられる特に、後者においてグルタミン酸シグナル伝達の欠損により骨粗懸症が発症することを発見した。
(4).VGLUTの非可逆的特異的阻害剤を発見した。
(5)ヌクレオチド(特にATP)のシグナル伝達に関わるトランスポーターを世界に先駆け発見し、その性質を解明した。
