Research Abstract

【背景と目的】Paternally expressed gene-1(Peg-1)(別名Mesoderm specific transcript (Mest))は、発生時に父親由来のアレルより発現するインプリンティング遺伝子である。我々は既に、肥満動物の肥大化脂肪細胞においてPeg1/Mest遺伝子発現が著しく増大することを見出した(Am. J. Physiol. 288: E117-E124, 2005)。本研究では、肥満の脂肪組織におけるインプリンティング遺伝子Peg1/Mestの発現制御を解析した。
【方法と結果】DNAデータベースサーチにより、Peg1/Mest遺伝子産物には、異なる第1エクソンによりコードされる複数のアイソフォームが存在することが明らかになった。real-time PCR法により、肥満の脂肪組織における発現誘導では、各アイソフォームに共通する第2エクソンの約2.3kb上流のゲノム上に位置する第1エクソンより転写開始されることが明らかになった。そして、Peg1/Mest遺伝子中に一塩基ポリモルフィズムを有するC57BL6マウスとJF1マウスを交配し、産仔を肥満させ脂肪組織で発現するPeg1/Mest cDNAの塩基配列を解析することにより、肥満の脂肪組織ではPeg1/Mest遺伝子が父方由来のアレルから転写されていることを見出した。【考察】インプリンティングの概念は個体発生時の遺伝子発現制御機構として位置付けられてきたが、本研究により、成体における肥満の脂肪組織でPeg1/Mestが片親性発現を維持したまま発現が増大することが明らかとなった。以上よりインプリンティング遺伝子Peg1/Mestは、胎仔成長以外にも脂肪細胞機能等、成体細胞の機能にも重要な役割を果たすことが示唆された。
