Research Abstract

本研究では、非競合的N-メチル-D-アスパラギン酸(NMDA)受容体拮抗薬のフェンシクリジン(PCP)連続投与マウスに認められる精神行動障害の発現機序の解明を目的とし、グリア型グルタミン酸トランスポーターの関与について検討を行った。PCP連続投与マウスの前頭前皮質では細胞外グルタミン酸遊離量の減少およびグリア型グルタミン酸トランスポーター(GLAST)の発現量の増加が認められた。PCP連続投与マウスの前頭前皮質にグルタミン酸トランスポーター阻害剤を注入したところ、前頭前皮質細胞外グルタミン酸遊離量が増加し、強制水泳における無動状態の増強(意欲低下の増強:情動障害)が緩解した。一方、GLAST遺伝子のヘテロ欠損マウスにPCPの連続投与を行ったところ、PCP連続投与野生型マウスに認められる精神行動障害が惹起されなかった。以上の結果よりPCP連続投与マウスに認められる精神行動障害の発現にはグリア型グルタミン酸トランスポーターの機能変化が関与すると示唆される。
