Research Abstract

我々はこれまでに、bHLH型転写因子Pancreatic transcription factor la (Ptfla)が、小脳原基脳室帯の放射状グリアにおいて発現し、小脳GABA作動性神経細胞の発生を司っていることを明らかにし、さらに菱脳唇放射状グリアで発現するもう一つのbHLH型転写因子Math1の知見と考え合わせることによって「異なる種類のbHLH型転写因子による放射状グリアの領域化モデル」を提唱してきている(Hoshino, The Cerebellum, 2006)。」本研究においては、後脳の放射状グリアの領域化とそこにおけるPtflaの働きについて調べた。
尾側後脳における放射状グリアは、背側からMath1, Ngn1, Ptfla, Ngn1というお互いに重なり合わない発現をするbHLH型転写因子によって領域分けできること、リニエージ解析からPtflaを発現する放射状グリアから下オリーブ核の登上線維神経細胞が生み出されることがわかった。また、Ptflaノックアウトマウスの解析から、Ptflaが登上線維神経細胞の運命決定、移動、分化、生存に関与していることが示唆された(J. Neurosci. 2007)。さらに現在は、蝸牛神経核を生み出す吻側後脳におけるPtflaおよびMath1の働きについても解析を進めている。
