Research Abstract

グリア細胞の細胞レベル・回路レベルでの機能について、最近の研究の進展は目覚ましいものがあるが、システムレベル、特に行動レベルでの機能については、その詳細はよくわかっていない。本研究では、グリア細胞に発現する遺伝子についての各種の遺伝子改変マウスの行動を、「網羅的行動テストバッテリー」を用いて解析することにより、各種のグリア関連遺伝子の行動レベルでの機能について明らかにしていくことが目的である。具体的には、本特定領域研究「グリアーニューロン回路網による情報処理機構の解明」の班員が作成した遺伝子改変マウスについて、「網羅的行動テストバッテリー」による解析の希望を募り解析支援を行った。研究代表者の研究室では、既に知覚・運動機能、情動行動、学習・記憶能力など、各種の行動の代表的なテストを集めバッテリー化した「網羅的テストバッテリー」がスムースに動いている。本研究では実際にこれを使用して、「グリア班」の班員らが作成したグリア関連遺伝子の遺伝子改変マウスの表現型解析を行う共同研究を行った。行動テストバッテリーによるデータ取得が既に一通り行われたものとしては(内は班員名、敬称略):MEL遺伝子欠損マウスの網羅的行動解析(瀬原淳子;論文準備中)、海馬交連切断単眼遮蔽マウスの空間学習特性についての研究(重本隆一)、GRPR欠失マウスとCaMKIIヘテロマウスと掛け合わせることによる表現型rescueの試み(和田圭司)、PLP欠失マウスの網羅的行動解析(池中一裕)、およびアレクサンダー病モデルマウスの網羅的行動解析(池中一裕)、14-3-3εヘテロ欠失マウスの網羅的行動解析(尾崎紀夫)、がある。このうち、海馬交連切断単眼遮蔽マウスの空間学習特性についての研究と14-3-3εヘテロ欠失マウスの網羅的行動解析については論文を投稿中である。PLP欠失マウスについては、社会的行動の低下、不安様行動の亢進、学習・記憶行動の障害など、統合失調症様の行動異常が見いだされ、オリゴデンドロサイトの異常が、統合失調症の症状の一部の原因になっている可能性が示唆された。
