Research Abstract

Opalinは、中枢神経系ミエリン膜に特異的に発現するタイプ1膜貫通型の糖タンパク質で、哺乳類ゲノムにのみ存在する系統進化学上新しい遺伝子である。タンパク質は、成熟オリゴデンドロサイトのミエリン形成期に発現が上昇して、免疫ゴールド電顕解析では主にパラノード・ループとインナー・ループの膜に局在する。短いアミノ末端領域にある2つのアスパラギンAsn-6とAsn-12には、シアル酸を含有したN型グルカンが付加される。細胞表面に存在するシアル酸は、カルボキシル基(負電荷)によって水分子(正電荷)と相互作用することにより、細胞認識や細胞膜間の距離等に関与する事が知られている。従って、Opalinの機能を知る上でグルカンの構造と機能の解析は重要である。今年度は、Thr-14にシアル酸含有O型グルカンが付加されることを明らかにした。また、加齢とともにOpalinの分子量が増加し、その増加する主成分はシアル酸であることを示した。さらに、培養細胞に形質発現したOpalinが細胞辺縁部と細胞突起の突端部に局在し、一方、N-グルカンとO-グルカン付加部位の欠失変異体は細胞内分布が変化することから、シアル酸含有グルカンがOpalinの細胞内輸送あるいはOpalinを介した細胞膜間の認識に何らかの作用をもつことが示唆された。ノックアウトマウスの作製に成功しており、さらに詳細な解析によってOpa1in機能を明らかにできるものと期待している。
