Research Abstract

1、ディスコイディンドメイン受容体2 DSドメインの繊維状コラーゲン認識様式の解明
NMRによりディスコイディンドメイン受容体2(DDR2)-DSドメインの立体構造を明らかにした。さらに、不溶性超分子である繊維状コラーゲンとの相互作用に当研究室で開発した転移交差飽和(TCS)法を適用し、DSドメイン上のコラーゲン結合部位を同定した。コラーゲン結合部位は、DSドメインのループ領域において、荷電性残基と疎水性残基から構成される溝を形成していることが明らかとなり、これまで構造生物学的解析が困難であった繊維状コラーゲン認識の特異性に関する分子基盤が得られた。
2、脂質二重膜中の膜蛋白質複合体における分子間相互作用解析法の確立
膜蛋白質は、従来、界面活性剤で可溶化して構造生物学的解析が行われてきたが、界面活性剤による可溶化はしばしば膜蛋白質の失活につながる。そこで、我々の研究室では膜蛋白質をアフィニティービーズに固定化した状態で脂質二重膜中に再構成するBead-linked proteoliposome(BPL)を開発した。しかし、BPLを用いたNMRによる分子間相互作用解析には固体の共存によるスペクトルの低感度化・低分解能化が問題となっていた。そこで、今回、マジック角回転によるスペクトルの高分解能化を試みた。まず、ユビキチン(Ub)と結合する酵母Ub水酸化酵素(YUH)の相互作用系において、YUHをヒスチジンタグを介してZn-NTAシリカビーズに固定化し、不均一系超分子を模倣した系を作成した。マジック角回転条件下で転移交差飽和法の測定を試みたところ、従来法では解析が困難であった固体と液体が共存した超分子系において、Ub上のYUH結合界面残基の同定に成功した。本手法は、膜蛋白質複合体における分子間相互作用解析に適用可能であり、今後の展開が期待できる。
